Doom, film diretto da Andrzej Bartkowiak, è ambientato a Olduvai, una stazione di Marte, dove le comunicazioni con la Terra si sono stranamente interrotte e uno degli ultimi messaggi giunti è un SOS. Gli scienziati presenti nella base sembrano essere stati attaccati da qualcosa di non ben identificato, che ha decimato molti dei presenti, lasciando i pochi sopravvissuti in quarantena e a rischio.
Per salvare quelli rimasti in vita ed eliminare la minaccia aliena, vengono inviati in missione sul pianeta un gruppo di marines, guidati dal comandante Sarge (Dwayne Johnson). Giunti a Olduvai, i soldati si imbattono nel responsabile della stazione, Markus Pinky (Dexter Fletcher), alcuni scienziati ancora vivi, tra cui il dottor Carmack (Robert Russel); in stato confusionale, e si scontrano con delle strane creature aliene, denominate Imp.
Il gruppo riesce a uccidere uno dei mostri e ad analizzarlo, scoprendo che sono in realtà parte dei scienziati ritenuti morti, trasformatisi invece in queste creature marziane a causa di un siero creato in laboratorio. I marines insieme alla dottoressa Samantha Grimm (Rosamund Pike) individuano anche il primo infetto, Hell Knight, che riesce a fuggire sulla Terra.
Per affrontare il mostro e i suoi nuovi adepti sul nostro pianeta la dottoressa Grimm decide di inniettare a suo fratello John (Karl Urban), uno dei marine, il siero, convinta che se somministrato a un uomo dal cuore buono, trasformi l'ospite in un superuomo e non in un mostro. La sua teoria sarà vincente oppure John diventerà una ripugnante creatura?
