Nel sud degli Stati Uniti, due anni prima della guerra di secessione, lo schiavo nero Django (Jamie Foxx) è riuscito a riacquistare la sua libertà grazie a King Schultz (Christoph Waltz), un ex dentista tedesco trasformatosi in cacciatore di taglie, che gli fa da mentore e guida con la speranza che Django lo conduca fino ai pericolosi fratelli Brittle. Dopo aver imparato a destreggiarsi tra pistole e duelli, Django diventa a sua volta un ottimo cacciatore di taglie ma in testa continua ad avere un solo obiettivo da concretizzare a qualunque costo: riuscire a rintracciare la moglie e liberarla dalla schiavitù a cui la costringe Calvin Candie (Leonardo DiCaprio), il diabolico proprietario di alcune piantagioni nel Mississippi, dove gli schiavi vengono allenati per combattere l'uno contro l'altro per il puro divertimento del loro padrone.
