Peter Parker, abbandonato bambino dai suoi genitori, cresce con gli zii Ben e May. Ormai liceale, non ha amici e s'innamora della compagna di scuola Gwen Stacy. Un giorno il giovane scopre una misteriosa valigetta che apparteneva al padre e decide di indagare sulla scomparsa dei suoi genitori. Le ricerche conducono alla Oscorp e al laboratorio del dottor Curt Connors, un ex collega di suo padre.
